Data preparation requirements

  • Find good, homogeneous, data; minimize interference
  • Capacity subtract transients if you are doing voltage jump

Assumptions

  • Data are Markovian - the channel can’t remember being in more than the last state (important, but easily satisfied)
  • The noise is white and Gaussian (not critical for most data)
  • The rate constants do not vary through a single data set. You can’t use time varying stimuli (but you can fit across sets of data obtained under different conditions). Time varying stimuli are supported only by Mac and MIP.
  • A significant fraction of events reach full amplitude - fast buzz model kinetics are (currently) out
